Q: Can I open a locked door to a room two roomates have vacated?

I am signed on a lease with these individuals for a year rental house, both vacated the premise, but still pay rent. However, they locked the door, which contains the second bathroom connected to the room. I asked the landlord if I could change the lock out, as none of the other doors have one, and he said yes.

A: If the roommates signed the lease and still pay their portion of the rent, it appears that the right to occupy the room still belongs to them. If you enter the room you would, in theory, be trespassing (absent some sort of emergency). I suggest you contact them and ask for permission.
