Q: Does throwing a knife, that falls short, and the thrower claims no intended harm still constitute assault?

A: It actually could be much worse then assault. Perhaps even throwing a deadly missle/object which is a 2nd degree felony. If you have been charge or are being investigated, you need an attorney immediately.

A: Yes it can. Assault is a threat to do harm, coupled with the present ability to carry out the threat. Here the knife was thrown, that completed the crime. The knife falling short does not eradicate the fact that it was thrown. Every defendant says he or she did not have the requisite intention.
